Our lab uses C. elegans as a model organism to study natural variation in response to stress. To conduct our research, we need many different maintenance media for the nematodes. One of those media is S-Medium, which requires a mixture of trace metals. Many labs purchase the metals individually, but US Biological sells a pre-mixed powder of the metals. Purchasing the pre-mixed powder allowed us to order fewer items and to weigh out fewer reagents when making the combined trace metals solution (rather than weighing multiple metals, we just weigh the pre-mixed powder).

Protocol Overview

To make the Trace Metals solution, we followed the directions provided by US Biological: "Dissolve 3.06g Trace Metals Mix into 1 liter of distilled/deionized water, heating with stirring until completely solubilized". We then aliquot the solution into light-blocking bottles and autoclave. We then use the solution to make S-Medium.

Results Summary

Overall, solubilizing the trace metals powder was easy. When I first attempted to dissolve the trace metals according to the provided documentation from US Biological, I had a problem getting everything to dissolve. I contacted the company and worked with a technical researcher. They were very responsive and helpful. The issue was solved.

Summary

The Good

It was much simpler to weigh out one reagent rather than 5 or so. Plus, it is cheaper to purchase the pre-mixed powder rather than the individual metals.
