Our work is in lysosomal degradation pathways and since we do a lot of lysosomal activity studies, in terms of chaperone-mediated autophagy, a specific antibody against hsc70 is a must. There are too many antibodies against heat-shock cognate (hsc) proteins and most of the antibodies against hsp70 also recognize hsp90 and hsp40. We have verified this antibody and it only recognizes hsc protein of 70kDa and no other hsc proteins. In addition, since hsc70 and other hsc proteins are also present as abundant cytosolic proteins, a specific antibody is a necessity to report if there are any changes in a particular scenario or condition. Great antibody.

Summary

The Good

Specific for hsc70, and very strong band at 70kDa

The Bad

Secondary is IgM, so have to invest in buying an IgM-HRP, which is somewhat rare to find in a general lab stock box.

The Bottom Line

Great antibody in terms of specificity and antigen-antibody binding. Being an abundant cytosolic protein, methanol fixation can be used in tracking down the specific antibody signals only in organelles and not the cytosolic hsc70 signal.
