The Alexa 488 from Molecular Probes is consistently good. I am on my third vial and am thrilled with the new packaging as it now comes in a light proof dark brown tube. This is great for those of us that have left a fluorophore out on the bench top, at RT, overnight before. For shame! Even before the new packaging, my terrible oversight in forgetting to store it properly, did not adversely affect the fluorophore at all. I went ahead and tested it, and it worked just fine. These are great antibodies.

Starting Material

Fixed third-instar Drosophila brain and antennal lobe complexes

Protocol Overview

4% paraformaldehyde fixation, 3x Horse Serum in PBS with Triton blocking, primary antibody 1:200 o/n at 4 degrees. 3x Horse Serum in PBS with Triton blocking/washes, secondary Alexa Fluor 488 1:200 at RT for an hour. Wash and mount in Slow Fade. Image using green laser.

Tips

No tips. Have good results O/N at 4 degrees or for 1 hour at RT.

Results Summary

Nice bright fluoresence. In my experience, fluoresence is maintained for at lease six months when sample is mounted in Slow Fade and slides are stored at 4 degrees.

I just overwhelmingly prefer Molecular Probes antibodies. No futzing around and they last forever.
