Company:

EMD Millipore

Product Name:

Anti-microtubule associated protein 2 (MAP2)

Catalog Number:

AB5622

Following MPTP, we wanted to quantify the total number of surviving neurons in the SNpc using an antibody specific for MAP2. Fixed brain tissue was sectioned through the midbrain containing the SN and stained (free floating) with anti-MAP2 and nuclei stained with DAPI containing medium. Stereological analysis was performed to quantify neuron numbers.

Sample

Fixed brain tissue

Primary Incubation

1:1000 in 2% normal donkey serum/TBS

Blocking Agent

5% NDS/TBS

Secondary Incubation

1:2000 in 2% NDS/TBS, 1 hour, room temp

Tertiary Incubation

Detection

Texas Red fluroescence

Results Summary

We were able to distinguish cell bodies from other staining but the images weren't fantastic (there was a lot of other staining in the tissues). But with the addition of the DAPI-containing medium we could pick out a cell confidently.

Summary

The Good

Easy to optimize concnetrations

The Bad

Doesn't result in publication-quality images

The Bottom Line

For what we use it for, this antibody works well enough that we will continue to order it.
