We are the histopathological service project in two Collaborative Research Centers and deal with various different mouse models of inflammation and infection Until now, the staining of mouse CD4 was only possible on cryo sections that are inferior to paraffin sections with respect to preservation of morphology and disinfection. With the anti-CD4 from eBioscience we are now able to stain CD4-positive cells in paraffin sections of mouse tissue. The best results are obtained after antigen retrieval using citrate buffer (pH 6). For detection, the LSAB method or immunofluorescence are suitable.

Applications

Immunohistochemistry

Sample

Paraffin sections of any mouse tissue

Primary Incubation

1:200, 30 minutes, room temperature

Blocking Agent

None

Secondary Incubation

Biotinylated rabbit anti-rat (1:1000), 30 minutes, room temperature

Tertiary Incubation

Streptavidin-AP, 30 minutes, room temperature

Detection

ABC from Dako

Results Summary

The best results were obtained with an antigen retrieval using citrate buffer though EDTA buffer is also applicable. The staining after EDTA retrieval is less crisp and has more background.
