The general aim of our research is to evaluate the role of a metabolic enzyme in generating host-protective Type 2 immunity against helminths. The specific goal of the experiment was to quantify IFNg secretion from myeloid cells deficient in the metabolic enzyme. We used this ELISA kit to quantify the IFNg concentration (pg/ml) from culture supernatants as well as bronchioalveolar lavage fluid. We selected this product for its ease of use and optimization. We actually use half the volumes recommended and it works well. The standards are great too.

We follow the manufacturer protocol. The plates are coated with capture antibody overnight. Then, the plate is washed and blocked for 2 hours at room temperature. The samples and standard curve are added and incubated overnight. On day three, the plate is washed and incubated with developing antibody for 1 hour. After another wash, TMB substrate solution is added for 30 minutes. The plate is washed and TMB solution is added. It takes just a few minutes to develop. The plate is stopped with acid solution and then read on an ELISA reader.
Wash plate 5x 200 ul evenly.
Very low background so the blanks are close to zero. Standard curves develop quickly. Samples develop quickly, very low detection. Highly concentrated samples need to be diluted. Data is reproducible and publishable because of the reliability and reproducibility of our ELISAs (particularly from one in vitro experiment to another).
