US biological G418 powder was used at 200mg/L in agar-containing YPD media for the selection of kanamycin-resistant S. cerevisiae. Yeast strains without the KanR gene failed to grow on this media, while strains with the resistance marker grew quite well.

Before pouring YPD plates, add 200mg/L G418.
Add the G418 to media as a powder without first dissolving in water.
KanR strains will grow on YPD + G418, while strains without the marker will not.
