This antibody was used to perform immunohistochemistry in paraffin-embedded tissue sections of mouse mammary gland and human breast tissues. Several antibodies against lysyl oxidase (LOX) were tested. The antibody reviewed here gave optimal staining in both mouse and human samples.

Primary Incubation

Primary antibody: Rabbit anti mouse, human, rat lox Millipore (Clone #ABT112) was incubated overnight at 4C at 1:200 dilution in TBS with 1% BSA

Blocking Agent

5% horse serum in TBS, 1% BSA for 2 hours at room temp with gentle rocking

Secondary Incubation

Horse anti-Rabbit Ig Vector ImmPRESS Reagent Kit (Cat # MP-7401) for 1 hour at room temp

Results Summary

This antibody was used to perform immunohistochemistry on paraffin sections of mouse mammary tissue (normal and tumor) and human breast tissue (normal and tumor). For each tissue type, the antibody was first tested over a range of dilutions including 1:50, 1:100, 1:200, 1:300. Parallel tissue sections were stained with secondary antibody alone. Staining of lysyl oxidase was observed for all dilutions tested. The dilution of 1:200 produced low background staining and retained easily detectable signal for positive stromal cells.
