We do total RNA sequencing to see all mRNA transcript this allows us to see whether particular genes are up or down compared to control treatment samples. For doing total RNA sequencing we need very high quality RNA. For this purpose we have been using Qiagen RNeasy mini kit to isolate RNA from tissue samples. We always get very high quality RNA.

After homogenizing the lung tissue we used the RNeasy column to bind RNA on it. Then, washing with buffer to get rid of all impurities, nuclease free water is used to elute the RNA from the column.
When you take RNA containing phase from the tube after centrifugation, be careful not to touch the middle layer with tips.
You can get 100% pure RNA with very high yield.
You can use this kit to isolate RNA from any kind of mouse tissue and cells.
