The microglial cell line, BV-2, is cultured every 3-4 days in a new medium in a T75 flask. These T75 flasks work great for the culturing of BV-2 cells. With the use of the T75 flask I am able to recover about 10-15 million cells after 3-4 days from just 1-2 million cells originally.

Protocol Overview

The BV-2 cells are lifted from an old T75 flask with the use of .01% EDTA. Cells are then centrifuged, counted, and replated at 1-2 million cells in 10mls of media in a new T75 flask.

Tips

To fully loosen the cells from the plate after the addition of EDTA, tap the plate against your hand or the table.

Results Summary

Summary

The Good

The flasks work extremely well with both adherent and non-adherent cells.

The Bad

Sometimes the cells require more than 20mls of media, and any more than that can cause the media to slosh up into the cap on the flask.

The Bottom Line

The T75 flask from BD Biosciences is a great tool to use for the culture of BV-2 cells, and really any type of cell you might have.
