After spleens have been removed and smashed though a 70 micron filter, the 18G needle is attached to a 5ml syringe, next the splenocyte mixture is drawn up very slowly into the syringe and expelled very slowly into another 70 micron fliter on a 50 ml tube.

Tips

Draw up and expel the mixture very slowly to prevent lysis of your cells of interest

Results Summary

With the use of two 70 micron filters and the 18G needle attached to a 5ml syringe, the spleen cells were broken down from the spleen into a single cell suspension with very little extra material (such as fat tissue) making it into the final cell suspension.
