Company:

Sigma Aldrich

Product Name:

Monoclonal Anti-β-Actin−Peroxidase antibody produced in mouse

Catalog Number:

A3854

Doing Western blots became a lot easier using this pre-conjugated beta-actin antibody. I use it on brain homogenate from MPTP-intoxicated mice to normalize protein concentrations loaded onto the gels.

Experimental Design and Results Summary

Applications

Western Blot

Sample

Brain homogenate

Primary Incubation

1:5000 in 5% dry milk/TBST

Blocking Agent

5% dry milk/TBST

Secondary Incubation

NA

Tertiary Incubation

NA

Detection

SuperSignal West Pico Chemiluminescent Substrate, 5 min room temp

Results Summary

This antibody is so nice to use since it is pre-conjugated so it saves time when doing Western blots. The bands are always very clear (I load 40 ug total protein onto my gels) and they show up at the correct molecular weight.

Summary

The Good

Saves time

The Bad

More expensive than unconjugated beta actin

The Bottom Line

Since I started using this antibody I don't think I'll go back to using un-conjugated beta-actin on Western blots. When you have to run a lot of Western blots, anywhere you can save time is nice.
