This product is used to transfer proteins from sds-page gel to PVDF membranes of 45kda and ~14 kda proteins. Compared to wet transfer it reduces the time (15min 20v), consumes less transfer buffer (40ml/gel from Thermo), and is more user friendly.

Protocol Overview

Keep the gel in milliQ water for 10 minutes, activate the PVDF with methanol for 1 minute and place in semidry trasfer along with whatman paper. Arrange gel, PVDF and whatman paper and set the voltage and time. (20v-15min)

Tips

Standardize the voltage and time as per the protein molecular weight which needs to be transferred.

Results Summary

It transfers the protein from SDS-PAGE to PVDF completely, but sometimes the bands diffuse due to excess heat and more amount buffer.
