Mouse calretinin antibody from Invitrogen was tried on mouse ovary tissue successfully. IHC staining on mouse tissues for this antibody was not reported in the data sheet. Mouse on mouse staining makes this experiment a little difficult. But blocking with goat serum as well as incubating the primary antibody with 1% serum helped to get a good outcome.

Experimental Design and Results Summary

Applications

Immunohistochemistry

Sample

Mouse ovary (control and Tumor)

Primary Incubation

1:200 dilution in 1% goat serum , overnight at 4 C

Blocking Agent

10% goat serum

Secondary Incubation

Biotinylated Goat anti mouse RTU secondary from Invitrogen, room temperature for 60 minutes

Tertiary Incubation

AEC Chromogen

Detection

Streptavidin HRP and AEC chromogen

Results Summary

Six months control ovarian surface epithelium expressed calretinin, stained red in the cytoplasm while the experimental tumor sample shows lack of expression and negative staining in its surface epithelium.

Additional Notes

Dysregulated estrogen receptor signaling in the hypothalamic-pituitary-ovarian axis leads to ovarian epithelial tumorigenesis in mice. Laws MJ, Kannan A, Pawar S, Haschek WM, Bagchi MK, Bagchi IC. PLoS Genet. 2014 Mar 6;10(3)

Summary

The Good

Good antibody for detection in mouse tissues.

The Bad

Mouse on mouse staining is always tricky. Background staining is difficult to avoid.

The Bottom Line

Overall, it is a good antibody to work with in both mouse and human tissue sections.
