I wanted to detect calretinin in fixed brain tissue via immunohistochemistry. I pulled this antibody out of a -20 degrees C freezer and tried to use it. It works very well while highly dilute and gives great pictures.

1% normal donkey serum in PBS (0.1% Triton-X 100)

1:1000 Cy3-conjugated donkey anti-goat for 2 hours at room temperature

Detection

Cy3 fluorescence via fluorescent confocal microscopy

Results Summary

Filled cells but images were crisp and clear. Antibody continues to work after being left for years in the -20 degree freezer.

Additional Notes

Did not show up for Ni-DAB stain with ABC kit until 1:100,000 dilution.
