The long-term goal of our lab is to understand the molecular and genetic mechanisms underlying brain development and diseases. My focus is studying the critical role of MeCP2 (methyl-CpG binding protein 2), which causes Rett Syndrome if mutated, in the development and function of the nervous system. I discovered a novel phosphorylation site on MeCP2 and plan to perform biochemical analysis in the N2a neuroblastoma cell line, therefore I want to study the distribution of total MeCP2 protein in the cell line first. To detect the distribution of MeCP2 in N2a blastoma cell line, N2a cells were immunostained with MeCP2 and DNA dye DAPI.
Immunofluorescence
Neuroblastoma N2a cell
1:2000 at 4°C for 12 hours
3% BSA and 0.1% Triton 100 in PBS buffer
1:1000 for 1 hour at 23° C

Confocal imaging
MeCP2 protein was expressed in the nuclei of N2a cells as punctas, which may represent CpG-rich regions that are repressed for transcription.

This antibody is good for immunostaining of a cell line.
