The detection of apoptosis and necrosis in cells is important in cancer research. Many labs, including mine, are working on developing new treatments for cancer. One of the first and most important steps in the development is to assess its ability to kill cancer cells. We chose this kit because of the combination of colors (APC and PI) which require minimal compensation on analysis and thus free up the FITC/GFP channel to use for when we use GFP+ cells.

The kit is simple. Just wash the cells and add the reagents! The annexin V binding buffer is provided, which is nice. There is even no need to wash the cells afterwards; they can be read immediately after staining (which is only 15 minutes at room temperature).
You can probably get away with using less of the reagents!
The results are usually displayed as a 4-quadrant dot-plot (see figure). The lower-left quadrant shows the percentage of viable cells; the lower-right quadrant shows the percentage of apoptotic cells; the upper-right quadrant shows the percentage of late-apoptotic/necrotic cells; and the upper-left quadrant shows the percentage of necrotic cells.
