We wanted to study phagocytosis by microglial cells. In our study we wanted to specifically treat cells with different stress inducers and observe the effect of such stress on microglial phagocytotic ability. For this experiment we used BV-2 and N9 mouse microglial cell line and plated them in poly-L lysine coated glass cover slips in 24 well plates. In the meantime, the pHrodo particles were mixed in 1X PBS (pH 7.4) and sonicated for 5 minutes at high frequency. 24 hours later the pHrodo particles were nicely vortexed and 20 µl of solution was added to each well. Phagocytosis was allowed for 2 hours and then cells were fixed and co-stained with hoechst dye.

Protocol Overview

Tips

Make sure the pH of the media is around 7 when the cells are cultured. For phagocytosis it is better to use the Live imaging solution from Life Technologies. Make sure to standardize the amount of beads to be added to get the best data.

Results Summary

The cells were imaged using Zeiss confocal microscope at 60X. The blue colour represents the nucleus of the cell and the red dots represents the E.Coli pHrodo Red particles in BV-2 cells .

The fluorescent bodies glow only when they are in the cells so background is less.

The E.coli particles stick to each other in media, thus you cannot specify the size of objects that the microglial cells are phagocytosizing

It is a very nice product and can be used to quantify data using different approaches.
