I am interested in molecular biomarkers for neurodegenerative conditions. I used this kit to detect albumin levels in human biofluids [plasma and cerebrospinal fluid]. I am familiar with this company and this particular kit was very well-reviewed and inexpensive. I am happy with the quality of the ELISA kit and will continue to purchase from this company. There is excellent intra- and inter-assay reliability and reproducibility.

Protocol Overview

Following desired sample dilutions and standard preparation, the samples/standards are added to the ELISA wells in duplicate. The plate incubates for 1 hour at room temperature and then is washed with wash buffer. Antibody is added to each well and the plate incubates for 30 minutes at room temperature. Following a second wash step, TMB substrate is added to each well and the plate incubates in the dark for 10 minutes at room temperature. Stop solution is added to each well and the colorimetric Abs is read on a plate reader at 450nm. Concentration can be calculated from the Abs readings.

Tips

I recommend shaking the plate on a microtiter plate shaker during the incubation steps.

Results Summary

The concentration of your samples are calculated from the standard curve. The samples and standards are run in duplicate alongside each other during the assay. The standard curve is best modeled with a four-parameter logistics equation. This can be done in Excel with the appropriate add-on option.

Albumin levels are generally high in human biofluids thus a large dilution is needed to fall within the detection range of the kit [this would be a similar problem with any albumin ELISA kit].

The Bottom Line

I have been very happy with this company and product. There is good inter- and intra-assay reliability and reproducibility. I will continue to purchase from this company for any ELISA kits I need for my research.
