We wanted an antibody to detect Pdx1, which is a transcription factor expressed in adult mouse islets and throughout the developing mouse pancreas.
Immunofluorescence
Adult mouse pancreas, fixed in 4 % PFA, frozen in OCT
1/100 in 10% FBS PBST, 4 C overnight
10% FBS diluted in PBST
Life Technologies Alexa 568; 1/200, 2 hours at room temperature or 4 C overnight (works either way); dilute in PBST
None used
Zeiss fluorescent microscope
This antibody works ok for detecting Pdx1 - similiar to or better than other available commercial antibodies. There is significant background though.

Works okay
Signal not very strong relative to background
I was unable to find another commercial antibody that worked any better than this one and that was approved for immunofluorescence.
