Useful myostatin ELISAs have been limited in the past. Our group is examining the endocrine responses of myostatin in healthy humans in response to atrophic stimuli. Plasma from healthy humans was run on this kit as standard and by manufacturers instructors.

Healthy human whole blood, spun for plasma

Protocol Overview

Exactly from manufacturers instructions, except for acidification step. For this key step, we found 40 uL HCl followed by 60 uL NaOH produced a more reliable result then the advised 50 / 50. As manufacturers state, end users need to check this step personally.

Tips

Do the acidification step, it's time consuming but necessary.

Results Summary

Results were excellent, with low coefficient of variability. Results were in a range of physiologically expected scores, and responded as hypothesized.

On attached image, lanes 1 - 3 show standard in triplicate, decreasing in concentration from A - H. Human plasma in all other wells

Does what's asked, cheaper than others on the market, most reliable that we've tested yet.
