When screening samples of varying composition, such as complex foods, obtaining a purified DNA template with fewer active inhibitors is advantageous over traditional template preparations such as boiling. This is why our lab often uses InstaGene to prepare DNA templates for amplification reactions such as PCR. The quick protocol and low cost of the reagent are also key factors making this a popular product.

Pure bacterial cultures or inoculated food samples

Protocol Overview

Isolate bacteria in a sample by centrifugation, add 200 ul of InstaGene matrix to the sample, incubate at 55 degrees C for 20 minutes, boil for 8 minutes, centrifuge and keep supernatant.

Tips

100 ul of matrix is often enough, and 2 ul of template is often sufficient for amplification.

Results Summary

Amplification is always observed if target bacteria is present in the sample, no contamination has been seen in InstaGene negative controls.
