Protocol Overview

The Life Technologies test assay is labeled with FAM dye, while the reference assay, which must be purchased separately, is VIC labelled. The assays are run in a duplex manner and the reference detects a target sequence known to be present in two copies in the diploid genome. Relative quantification is then calculated using the Life technologies CopyCaller software, which is freely available. Practically, the test and reference assay are combined with 5ng of DNA and genotyping master mix, then run as per a standard qRT-PCR assay. The protocol is very quick to perform and complete within 2-4 hours.

The reference assays must be validated

Results Summary

The results are adequate for validation of CNV analysis by other methods, as well as for quick visualization of CNV estimation.
