We wanted to perform qPCR testing to determine if treatment of a G-protein-coupled receptor with an agonist would affect the transcription of different genes. This machine worked well for us over multiple projects and generated a lot of data.

cDNA from tissue extracts and cell culture extracts

Protocol Overview

Tissue or cells were collected, treated with RNAlater(TM) and stored at -20 degrees C until extraction. RNA was extracted via TRIzol and the RNA was reverse-transcribed using a Superscript 3 kit.

Tips

Does not have a gradient function; must test annealing temperatures in separate experiments.

Results Summary

The machine worked well and consistently generated quality data, even when on for days straight. Plates were run every 2.5 hours for 24 hours, and none of the data from the plates showed any drift. The current software does a good job of identifying the right Ct, though you should still check if it's correct.
