I am interested in the expression of genes in the urine of transplant patients, in order to identify biomarkers for a disease called graft versus host disease. I have used the kit to isolate total RNA from urine samples in order to assess gene expression by downstream qRT-PCR.

The protocol is very straightforward to perform. Briefly, to each urine sample the user must add lysis buffer, slurry reagent and ethanol and cetrifuge at 2,000g. The supernatant is discarded, wash buffer added and the sample transferred to a spin column. Additional wash buffer is added and the column centrifuged. This wash step is repeated followed by a wash with ethanol and the sample then incubated at 60C for 3 minutes. Finally, total RNA is eluted by addition of elution solution and centrifugation.
Be sure to prepare a heat block to the correct temperature prior to starting the protocol.
We get good RNA recovery with sufficient quantity to assess gene expression by downstream TaqMan qRT-PCR.
