We study the various mechanisms of gene silencing responsible for disease resistance in poultry. We specifically study various target genes through genotyping. In addition to the high yields of PCR products, obtaining a pure template is pivotal for many downstream applications such as sequencing, ligations, in vitro transcription etc. We use the Minelute PCR purification kit to remove primers, salts and other reagents from the PCR reaction.

The manual elucidates the application of all the buffers used in the kit. The manufacturer supplies all the reagents required for the purification procedure. The pH indicator (color) of the binding buffer is critical and should be checked before starting the purification procedure. The protocol overview is as follows: to the template DNA add the binding buffer, spin and wash using the wash buffers, and finally elute with TE/H2O.
Including an additional centrifugation step after the wash steps will help remove any residual ethanol that could affect some downstream applications.
The yields of the purified product correlates with the manufacturers mentioned yield range.
The sample loss using the kit is low when compared to other purification kits.
