We overexpress IL-27 in the mice for determining its role in osteoclastogenesis. Mice serum are collected to check for the circulating IL-27 heterodimer levels. Utilizing the components in the biolegend kit, I can determine the levels easily in approximately 3 hrs and since plates are pre-coated I do not need to worry about coating the plates the previous night. I can extract the serum the same day and check fresh without worrying about protein degradation at all. The sensitivity is good and the background less. Perfect as always. No other company gives the same product with so less background and great result. Extremely happy with it. Will be ordering more in the future.

Protocol Overview

The kit was utilized per instructions in the manual. The serum sample when overexpressed was used at 1:100 dilution and when just from naive mouse used at 1:1 diution.

Tips

Not really. It is very simple. However, the only disadvantage is the frame that comes along with the wells. If you want to use few wells only, it is required to remove all the other wells as they are pre-coated and the wells do not fit any other frames such as from R&D systems

Results Summary

Results were generated against the standard provided and produced a straight line against log-log parameter which works really the best I figured for this kit.

Nice kit and works very well. Biolegend is doing a great job. Hope they continue producing similar products. I am looking for some great multiplex kit from them.
