Our research is in Animal, Dairy and Veterinary Sciences at Utah State University.

Sample collection and DNA extraction are the first steps for most experiments. Initial sample quality determines the quality of the end result. The general aim of the project is to study epigenetic regulation of genes responsible for metabolism of toxins. I use the kit routinely to extract gDNA from turkey liver or blood samples for SNP genotyping. The kit offers simple, easy and efficient extraction of gDNA from samples.

Protocol Overview

The kit provides all the reagents and tubes for isolating the gDNA, except PBS. The protocol is very well written and incorporates specific important notes describing the handling of reagents. A brief overview of the protocol is as follows: add appropriate amount of ProteinaseK to the sample followed by addition of PBS and buffer AL. Incubate samples at 56 C for 10minutes and add 95% ethanol. Wash with buffer AWI and AW2 in two separate steps. Elute the DNA in buffer AE/TE or distilled water.

Tips

If dealing with large number of samples, aliquoting proteinase K ahead will save time.

Additional Notes

Change of collection tube after each wash step is redundant. I would recommend discarding/dumping off the wash buffer after the spin to reduce the load of plastic waste.
