Our research lab is a HIV lab and we study how HIV affects the immune system and takes over the host cells. We use Jurkat cells and mouse thymocytes as model systems as well as patient samples. Performing a ChIp with patient samples can be a tough task. And if you perform ChIp in batches, the results may vary. That's when our lab bought an IPStar automated ChIp workstation.

Experimental Design and Results Summary

Application

ChIP for various human samples

Starting Material

PBMCs isolated from blood

Protocol Overview

ChIP was performed for various HIV proteins in samples collected from patients.

Tips

Simulate the whole program and check it twice. Smallest mistake can ruin the whole experiment and cost a lot of money.

Results Summary

The machine finishes multiple ChIPs in an overnight run. All one needs to do is put the chromatin, antibodies, beads and all buffers in designated reservoirs, set up the experiment via the software on connected laptop and run. The machine will perform all the steps and one has the immunopreciptated chromatin ready for downstream processing.

Features Summary

Everything is automated from the point you keep everything at designated places, reproducibility.

Additional Notes

Make sure you have kept sufficient number of tips in required holders. If you run out of tips midway, it can compromise the experiment

Summary

The Good

Total automation, computer controlled, simple software

The Bad

Expensive reagents (Diagenode advises to buy everything from them, even antibodies), machine is quite large in size, software is a bit confusing, expensive

The Bottom Line

A very good instrument if you are handling multiple ChIP samples and if you are concerned about the variations in different samples.
