I am using the Cytocell FLI1/EWSR1 probe to assess gain or loss of these target regions in lymphoma samples.  The probe can also be used to detect translocations between the two regions, as indicated by duel fusion yellow signal.

Experimental Design and Results Summary

Application

FISH for gain and loss

Starting Material

Fresh-frozen imprint slides

Protocol Overview

The probe was applied directly to the slides, which were incubated for 75C for 2 minutes followed by 37C for 24 hours.  Slides were then washed in 0.4xSSC with tween followed by 2xSSC and DAPI applied. The EWSR1 probe is labeled in green and the ATM probe is labeled in red. The probe can be used to look at gain or loss of these regions as well as translocations, as indicated by duel fusion (yellow).

Tips

DAPI is also supplied

Results Summary

Clear red and green signals, with two copies of each per cell indicating normal status
