Our lab studies the mechanisms of Aflatoxin metabolism through enzymes such as Glutathione S Transferases in poultry. We routinely use AmpliTaq Gold 360 DNA polymerase to amplify and validate SNPs on target genes (or regions) from various samples. The enzyme is also used in a PCR based method for early gender identification in turkey poults. As mentioned at the manufacturer’s webpage, the ultra-pure enzyme is optimized for varying PCR conditions. The PCR reaction can be set up at room temperature without concern for extension at misprimed sites. The PCR using the enzyme resulted in high quality sequencing data

The kit comes with different components, enzyme, 10x buffer, MgCl2 and GC enhancer. Mix the different components of the reaction at optimal concentrations/as recommended by the manufacturer. Add the Taq at the end. Mix well.
Follow manufacturer’s guidelines while optimizing PCR conditions. Include GC enhancer for difficult- to- amplify targets.
The PCR using the enzyme mostly resulted in high yield of amplified products with reduced non-specificity.
