Krt5 was used as an epithelial marker in mouse embryonic limb development and digit separation. Limbs were sectioned at 2um and immunofluorescent stained for Krt5 using anti-Rabbit AlexaFluor 546 secondary antibody.

Primary Incubation

1:1000, incubated overnight at 4 degrees C, diluted in DAKO antibody diluent

Blocking Agent

DAKO serum-free protein block + 5% Normal Goat Serum

Secondary Incubation

1:2000 Goat anti-Rabbit AlexaFluor 546 (2mg/ml), 2 hours at Room Temp, diluted in DAKO antibody diluent

Tertiary Incubation

Detection

Fluorescent microscope

Results Summary

Krt5 epithelial cells were very clearly labelled along the epidermal edge of the mouse paw. Background staining was almost completely absent.

Additional Notes

DAPI used to stain nuclei. We have not used this antibody for any other applications.
