My research work involved transfecting PC12 and differentiated PC12 cells to study the peripheral neuropathy after cisplatin treatments. I have to treat my cells with cisplatin after transfections with Sirt2 KD and overexpressed proteins. When using chemical transfections, the efficiency was very poor in differentiated cells. This kit works really nicely. Though they didn't mention that it could be used for differentiated cells, I just gave a try.  I saw no cell death and more that 60% transfection efficiency.

Protocol Overview

Just follow the kit protocol. It is very simple.  If doing for the first time, perform a test experiment in your cells before planning the actual experiment.

Tips

Sometimes we had high death rate of cells.  Therefore make sure cells are in growing phase during transfection.

Results Summary

I got 60-70% transfection efficiency even in primary cells
