ExoQuick serum exosome precipitation solution

I have used the ExoQuick solution to isolate exosomes from serum for subsequent downstream RNA extraction and qRT-PCR analysis. I previously compared ultracentrifugation methodology and found ExoQuick to give superior recovery in a fraction of the time and without the need for specialist equipment.
Isolation of serum exosomes
200ul serum
The protocol is simple and fast to perform. I started with 200ul of serum, which was centrifuged at 3000xg to remove cells and cell debris. The supernatant was then transferred to a new tube and 63ul ExoQuick added, mixed and the sample incubated at 4C for 30 minutes. Following this period, the samples were centrifuged at 13,000RPM for 2 minutes to pellet the exosomes. The supernatant was then removed and the exosome pellet can be re-suspended in water for additional analysis or lysis buffer directly applied if extracting nucleic acid.
The product is also available with thrombin for plasma, or as ExoQuick TC for use with urine or media.
The exosome pellet is clearly visible after centrifugation from as little as 200ul of serum. I have assessed the exosomes via electron microscopy and the appropriate sized vesicles are present. RNA can be effectively isolated by subsequent extraction and successfully analysed by qRT-PCR. The results were superior and faster to obtain than traditional ultracentrifugation methods.
