Urea is a key major end product of protein metabolism in humans. It constitutes the largest fraction of non-protein nitrogen component of the blood. Urea is produced in liver and excreted through kidneys in the urine. Consequently the calculating level of urea depend upon protein intake, protein catabolism and kidney function. I was in a search of a highly specific and sensitive protocol for estimation of urea as I am working on phytochemicals in the area of kidney stone research and I wanted to analyze effect of phytochemicals on the extent of stone formation as kidney function is mainly impaired due to kidney stone. I chose above kit for its sensitivity and reproduciblity

Protocol Overview

The principle of the urea Assay Kit is urea is converted into glutamate in a two step reaction with help of enzymes urease and glutamate dehydrogenase. The rate of decrease in absorbance is monitored at 340nm and is directly proportional to urea concentration in the sample.

Tips

The kit is sensitive enough to allow estimation in range of 250mg/dl levels and for higher values samples should be diluted with normal saline and results can be calculated by multiplying with dilution factor.

The results I obtained are accurate and comparable to earlier studies.
