Macherey Nagel TriPrep kit is intended to extract DNA/RNA/protein in a single protocol. This is highly needed and desired to get consistent results at all three levels.Extracting DNA and RNA from separate tissue parts, cell passages can result in unexplained variance. The kit is fairly easy to use, it has the maximum limit of 30mg of animal tissue and 100 mg of plant's. But the lower limit is not mentioned and the yield is not proportional to the weight of the sample. It depends on many other factors and most importantly homogenization. A thorough homogenization is needed to ensure good quality results.The DNA/RNA extracted from this kit are good for microarray applications. I used them for Affymetrix cytoscan HD and exon arrays.

Results Summary

The results are very good for both DNA and RNA but are not very predictable for old and low amount samples. Standardization needs to be done at the homogenization level. We used this kit for extracting DNA/RNA/Protein from biopsies and surgical resections. The range of DNA extracted was 0-70ng/ul with 100ul total volume. RNA was extracted in the range of 5-600ng/ul with 60ul total volume.Both DNA and RNA worked fine for microarray experiments.
