I am interested in the expression of microRNAs as biomarkers for onset and severity of graft versus host disease (GvHD). I frequently measure microRNA expression using qRT-PCR in the body fluids of patients with GvHD.

Starting Material

cDNA reverse transcribed from the total RNA of whole blood samples.

Protocol Overview

The Bioline qRT-PCR master mix is designed to be comparable to the Life Technologies TaqMan master mix. Thus, the protocols are comparable and each TaqMan RT-PCR reaction incorporates master mix, TaqMan Assay, cDNA and water. The master mix itself is made up from an optimised cocktail of heat-activated DNA polymerase, ultra-pure dNTPs, MgCl2 and can be purchased with or without ROX Internal reference

Tips

The master mix must be stored frozen

Results Summary

The results are a high performance reaction in terms of sensitivity and florescence. The amplification curves are good and the efficiency maximal. In our laboratory we observe lower Ct values using the Bioline master mix compared to Life Technologies.
