I needed to identify oligodendroglial cells in mouse brain for stereological analysis. Olig2 is a marker for oligodendroglial cells at all stages of development from immature oligodendrocyte precursors to mature myelinating oligodendrocytes. Also since Olig2 is a transcription factor it produces a nuclear staining pattern which allows for relatively easy cell counting.
Immunofluorescence
50 um thick, floating, postnatal mouse brain fixed in 4% PFA overnight at 4C
1:2000 dilution in blocking agent, overnight incubation at 4C
5% normal goat serum + 0.5% Triton X100
1:1000 dilution of secondary antibody in PBS, overnight incubation at 4C

Procedure results in nuclear staining of oligodendroglial cells throughout the tissue which can be visualized via fluorescent microscopy.
Works well at a relatively dilute amounts
Long incubation periods are needed for use at the dilute concentrations used.
Great antibody for immunostaining oligodendroglia in brain tissue.
