Tested Aviva Olig1 antibody to identify oligodendrocytes on mouse brain paraffin sections

Experimental Design and Results Summary

Applications

Immunohistochemistry

Sample

Paraffin-embedded formalin-fixed mouse brain section

Primary Incubation

1:100 in TTBS, 4 degrees, overnight

Blocking Agent

10% normal serum with 1% BSA in TBS

Secondary Incubation

Sigma Anti-rabbit #31460, 1:300, 1 hour, room temp

Tertiary Incubation

N/A

Detection

VectorLabs Novared, counterstained with hematoxylin

Results Summary

Staining seen with primary antibody, not with no primary control. Predominant signal consistent with oligodendrocyte nuclei (strongest in cerebellar white matter, pons and thalamus). Saw some signal in CA3 of hippocampus that I'm not sure should be there.Note: used antigen retrieval: 10mM sodium citrate, .05% Teen-20, pH6.0. Boiled gently for 20 minutes.

Summary

The Good

Strong signal using basic protocol

The Bad

Might have given some off-target signal, not sure

The Bottom Line

Antibody worked well but may need to costain with another oligodendrocyte marker to verify specificity before using as oligodendrocyte marker.
