PTEN is a tumor suppressor implicated in a wide variety of human cancers. It is a major negative regulator of the PI3K/Akt signaling pathway and also regulates p53 protein levels and activity. Furthermore, PTEN has been implicated in the pathobiology of diffuse large B-cell lymphoma. I am using the PTEN antibody to measure PTEN protein levels by immunohistochemistry (IHC) in diffuse large B-cell lymphoma formalin fixed paraffin embedded tissue sections.

Rabbit monoclonal (D4.3) antibody to PTEN (cat # 9188). The antibody was used at a 1/25 dilution in Ventana® Diluent. The protocol comprised a standard CC1 (Ventana®) antigen retrieval , 60 minute antibody incubation, 8 minute hematoxylin and 8 minute bluing reagent (Ventana®) in conjunction with the Ventana® ultraView™ DAB Detection Kit.
No blocking agent is required using this antibody with the Ventana® ultraView™ DAB Detection Kit.
