PTEN (D4.3) IHC staining in diffuse large B-cell lymphoma cells. The antibody demonstrates cytoplasmic and nuclear staining of large CD20-positive malignant cells.

Company:

Cell Signaling Technology

Product Name:

PTEN XP® Rabbit Monoclonal Antibody (D4.3)

Catalog Number:

9188

PTEN is a tumor suppressor implicated in a wide variety of human cancers. It is a major negative regulator of the PI3K/Akt signaling pathway and also regulates p53 protein levels and activity. Furthermore, PTEN has been implicated in the pathobiology of diffuse large B-cell lymphoma. I am using the PTEN antibody to measure PTEN protein levels by immunohistochemistry (IHC) in diffuse large B-cell lymphoma formalin fixed paraffin embedded tissue sections.

Experimental Design and Results Summary

Applications

IHC

Sample

4 micron sections cut from diffuse large B-cell lymphoma tumors

Primary Incubation

Rabbit monoclonal (D4.3) antibody to PTEN (cat # 9188). The antibody was used at a 1/25 dilution in Ventana® Diluent. The protocol comprised a standard CC1 (Ventana®) antigen retrieval , 60 minute antibody incubation, 8 minute hematoxylin and 8 minute bluing reagent (Ventana®) in conjunction with the Ventana® ultraView™ DAB Detection Kit.

Blocking Agent

No blocking agent is required using this antibody with the Ventana® ultraView™ DAB Detection Kit.

Secondary Incubation

Universal HRP Multimer containing HRP labeled antibodies; goat anti-mouse IgG, goat anti-mouse IgM and goat anti-rabbit (Ventana® ultraView™ DAB Detection Kit).

Tertiary Incubation

N/A

Detection

DAB (3, 3’-diaminobenzidine tetrahydrochloride)

Results Summary

The optimized staining conditions give specific cytoplasmic and nuclear staining.

Additional Notes

This antibody is from the XP® range, which is exclusively available from Cell Signaling Technology and has been carefully selected based on superior results. The XP® antibodies are generated using XMT® technology, which provides access to a broad range of antibodies unattainable with traditional monoclonal technologies.

Summary

The Good

Good value for money with specific staining.

The Bad

It can be challenging to distinguish between nuclear and cytoplasmic staining during cell scoring.

The Bottom Line

Good all-round antibody with specific results.
