Applications

Immunocytochemistry for myelination cultures from embryonic rat brain

Sample

A dissociated neuron-oligodendrocyrte co-culture was prepared from embryonic day 16 rat forebrain and cultured for 40 days. Cells were fixed with 4% paraformaldehyde for 10 min and then rinsed with PBS.

Primary Incubation

Anti-Caspr, used at 1:300, diluted in PBS containing 10% goat serum, incubated overnight at 4°C.

Blocking Agent

10% normal goat serum; blocked at room temperature (RT) for 1 hour.

Secondary Incubation

Biotin-labeled anti-rabbit IgG (from JacksonImmunoResearch, cat# 111-065-003), diluted at 1:250 in PBS containing 10% Goat serum. Incubated at RT for 1 hr.

Tertiary Incubation

N/A

Detection

Immunofluorescence. Using Avidin-Alex488 (Life Technologies, cat# S11223). Observed using a Olympus BX60 fluorescence microscope.

Results Summary

Using antibodies against Caspr and MBP for double-staining, the Nodes of Ranvier were revealed. This antibody produced strong positive signals with acceptable background.

Additional Notes

Also works fine in frozen rat brain sections.
