Our goal is to verify expression changes in several genes involved in interferon pathways, such as USP18, following compound treatment. We selected the 4813 antibody from CST because we’ve had good results with other antibodies from that supplier in the past.
Western Blot
Human immortalized cell line, whole cell extracts.
Primary Antibody: USP18 (D4E7) Rabbit mAb, Cell Signaling Technology, Cat # 4813. Overnight incubation at 4°C. We use this antibody at a 1:1000 dilution.
1x TBS, 0.1% Tween-20 with 5% w/v nonfat dry milk; 1 hr block at RT.
Goat anti-rabbit, Santa-Cruz sc-2004, HRP conjugated; 1 hr incubation at RT.

Our results with this antibody have been very clear, we see a very strong doublet (as expected) at the expected molecular weight, with minimal/no background signal.
