The site-directed mutagenesis service of GENEART introduces single or multiple mutations such as substitutions, insertions, deletions or truncations into your DNA sequence within shortest turnaround times. Construct will be completely sequence verified and are free of additional backbone mutations since no quick-quick change procedure is involved.
Deliverables All gene variants are subcloned into the customer's vector and delivered as 10 µg of plasmid DNA.
Depending on the details of the customer's screening assay, site-saturated constructs can be delivered in two different formats: *mixed (small library containing all 20 variants) *separate (each of the 19 substitution variants will be delivered separately)
Quality control *A mixed site-saturated library will be bulk sequenced. *Separated variants will be 100 % sequence verified each.
In vitro site directed mutagenesis is very useful for studying protein structure function relationships and for identifying intra-molecular regions or amino acids involved in gene expression. We can perform site-specific mutagenesis in any double stranded plasmid. Mutation can be: point mutations, switch amino acids, deletion, or insertion of single amino acids.
