Custom made highly complex, domain libraries

  • Description: Our libraries are the most complex ones available on the market and ULTImate Y2H™ cell-to-cell mating is the only screening technology that can screen them exhaustively. Should you not find your favourite library among our 60+ libraries, we would be happy to construct it for you.

    Our ULTImate Y2H domain libraries are validated by extremely stringent quality controls:

    • Verifying a complexity of at least 10 million independent, primary fragments in yeast for Random-Primed cDNA libraries. The complexity of the genomic libraries (constructed for organisms that contain no or few introns) depends on the size of the genome (e.g. 2 million genomic fragments for Helicobacter pylori library)
    • Random sequencing of 192 clones to check for i) absence of contamination, ii) that ribosomal RNA, mitochondrial DNA and empty vectors represent less than 10% of the total sequences
    • Verifying the viability and mating properties of the library-containing yeast cells, and defining the conditions to obtain the highest mating efficiency (Patent US No 6,187,535)
    • Reproducing selected Y2H screen interactions available from the literature.
