Description: In vivo siRNA delivery and tissue-targeting RNAi-induced gene silencing is used to study gene function in cultured cells, however this approach does not allow a critical evaluation of gene function within the whole organism. Unfortunately, application of RNAi in vivo may present many technical challenges due to siRNA stability, delivery efficiency, and tissue targeting.

Altogen Labs developed methods to prevent siRNA degradation in serum, increase delivery efficiency to target organ, therefore providing strong functional effect (phenotype) induced by siRNA in vivo.

There are 2 basic methods adapted for RNAi use in vivo: administration of small interferring RNA (siRNA), and administration of plasmid DNA and viral vectors, which express a short hairpin RNA (shRNA) that is subsequently processed into active siRNA in vivo. In general, viral-based shRNA vectors provide highest delivery efficiency in vivo, but unfortunately they are time-consuming to construct and have significant immunogenicity problem (as well as most retrovirus-, adenovirus-, and lentivirus-based delivery systems.
