pUNO-mMyD88
InvivoGen
Catalog Number
: puno-mmyd88
Description
: pUNO-mMyD88 is a sequence-verified expression plasmid containing the murine Myeloid Differentiation primary response gene 88 Myeloid differentiation factor 88 (MyD88) is a cytoplasmic adaptor protein that is involved in interleukin-1 receptor (IL-1R)- and Toll-like receptor (TLR)-induced activation of NF-kappaB. MyD88 contains a C-terminal Toll/IL-1R homology (TIR) domain and an N-terminal death domain (DD), which mediate the interaction of MyD88 with IL-1R/TLR and IRAK, respectively. MyD88 is essential for signaling as MyD88 -/- mice fail to induce cytokines in response to LPS and CpG DNA. Some TLRs induce antiviral responses by producing IFN-alpha in a MyD88-dependent manner. MyD88 interacts with IRF7 resulting in the activation of IFN-alpha-dependent promoters.1. Janssens S. et al., 2002. Regulation of interleukin-1- and lipopolysaccharide-induced NF-kappaB activation by alternative splicing of MyD88. Curr Biol. 12(6):467-71.2. Kawai T. et al. 1999. Unresponsiveness of MyD88-deficient mice to endotoxin. Immunity 11(1):115-22.3. Kawai T. et al., 2004. Interferon-alpha induction through Toll-like receptors involves a direct interaction of IRF7 with MyD88 and TRAF6. Nat Immunol. 5(10):1061-8. Plasmid is selectable in both mammalian cells and E. coli with Blasticidin.

pUNO-mSARM1b
InvivoGen
Catalog Number
: puno-msarm1b
Description
: pUNO-mSARM1b is a sequence-verified expression plasmid containing the murine Sterile alpha and HEAT-Armadillo motifs SARM1 is a putative new adaptor, described in 2001 as sterile a and HEAT-Armadillo motifs. SARM contains a TIR domain, two sterile a motif (SAM) and an Armadillo repeat motif (ARM). Both motifs are known to be involved in protein-protein interactions promoting the formation of diverse protein complexes. Thus, SARM possesses the characteristics of a signaling molecule but its role in TLR signaling is still unknown. Plasmid is selectable in both mammalian cells and E. coli with Blasticidin.

pUNO-mTIRAP
InvivoGen
Catalog Number
: puno-mtirap
Description
: pUNO-mTIRAP is a sequence-verified expression plasmid containing the murine TIR domain-containing adapter protein (TIRAP) TIR domain-containing adapter protein (TIRAP), also known as Mal, is differentially involved in the signaling to NF-kB by members of the TLR family. TIRAP-deficient mice were shown to respond normally to the TLR3, TLR5, TLR7 and TLR9 ligands, but to be defective in cytokine response to LPS, a ligand for TLR4, as well as ligands for TLR2, TLR1 and TLR6[1,2]. TIRAP contributes to TLR2 and TLR4 MyD88-dependent signaling pathway but interacts only with TLR4 to activate a MyD88-independent pathway to induce IFN-β and STAT-1 dependent genes. Plasmid is selectable in both mammalian cells and E. coli with Blasticidin.

pUNO-mTRAM
InvivoGen
Catalog Number
: puno-mtram
Description
: pUNO-mTRAM is a sequence-verified expression plasmid containing the murine TRIF-related adapter molecule TRIF-related adaptor molecule (TRAM), also known as TIR-containing adaptor molecule (TICAM)-2, is a recently characterized adaptor molecule involved in the MyD88-independent TLR4 signaling pathway. In response to LPS stimulation, TRAM physically bridges TLR4 and TRIF transmitting LPS-induced signaling to TRIF which in turn activates IRF-3 and the production of interferon beta. TRAM function is restricted to the TLR4 pathway, as TRAM-deficient mice are defective in cytokine production in response to TLR4 ligand but not to other TLR ligands.1. Fitzgerald KA. et al., 2003. LPS-TLR4 Signaling to IRF-3/7 and NF-{kappa}B Involves the Toll Adapters TRAM and TRIF. J Exp Med. 198(7):1043-1055. 2. Oshiumi H. et al., 2003. TIR-containing adapter molecule (TICAM)-2, a bridging adapter recruiting to toll-like receptor 4 TICAM-1 that induces interferon-beta. J Biol Chem. 278(50):49751-62 3. Yamamoto M. et al., 2003. TRAM is specifically involved in the Toll-like receptor 4-mediated MyD88-independent signaling pathway. Nat Immunol. 4(11):1144-50 Plasmid is selectable in both mammalian cells and E. coli with Blasticidin.

pUNO-mTRIF
InvivoGen
Catalog Number
: puno-mtrif
Description
: pUNO-mTRIF is a sequence-verified expression plasmid containing the murine TIR domain-containing adapter inducing IFNbeta TIR domain-containing adapter inducing IFN-b (TRIF), also named TIR-containing adaptor molecule (TICAM)-1, is a novel adapter involved in the MyD88-independent signaling pathway of several TLRs. A dominant-negative form of TRIF was shown to completely abolish TLR3-induced signaling and also to block the NF-kB activity mediated by TLR2, TLR4 and TLR7. Further analysis has revealed that TRIF binds preferentially to TLR3 and TLR4 and is essential in their signaling pathways. It predominantly activates IRF-3 and the production of IFN-b rather than NF-kB. 1. Yamamoto M. et al. 2002. Cutting edge: A novel Toll/Il-1 receptor domain-containing adapter that preferentially activates the IFN-b promoter in the Toll-like receptor signaling. J Immunol, 169(12):6668-66722. Yamamoto M. et al., 2003. Role of adaptor TRIF in the MyD88-independent toll-like receptor signaling pathway. Science. 301(5633):640-3. 3. Oshiumi H. et al., 2003. TICAM-1, an adaptor molecule that participates in Toll-like receptor 3-mediated interferon-beta induction.Nat Immunol. 4(2):161-7. Plasmid is selectable in both mammalian cells and E. coli with Blasticidin.
