    Using 32 different human tissues and 34 unique human cancer cell lines we constructed libraries for groups of cell lines and tissues and then pooled the primary clones to create the most comprehensive collection of the human transcriptome to date. Through careful library construction thereby preserving rare sequences and by a unique sizing procedure for larger molecules, we enriched for full-length cDNAs greater than 800 bp. Additionally, multiple tissues and cell lines sources used in this library allow for the discovery of alternatively spliced mRNAs differentially expressed in tissues. Thus, this collection maximizes diversity and rare genetic sequences.

    Uniquely Constructed Library Makes Cloning Genes Easy
    The MegaMan™ Human Transcriptome Library is a complex mixture of high-quality plasmid libraries representing tens of millions of primary clones. We constructed these libraries from diverse mRNA sources after enriching for the longest cDNA transcripts. The DNA samples were subsequently pooled with a ratio that maximizes the probability of good representation of even the rarest and longest transcripts. This is the most comprehensive template representing the human transcriptome and can be used for an extensive variety of cloning projects. It is ideal for cloning any well-characterized genes or transcripts previously identified by expressed sequence tag (EST) studies, as well as, for the validation of predicted genes and alternative splice isoforms generated from the Human Genome sequencing data.

    Cloning Methodology Delivers Only cDNA Transcripts
    The cloning technique used to create these libraries is extremely efficient for directional cloning of full-length cDNAs without blunting and ligating linkers onto the ends of the cDNA molecules (Figure 1A). As a result, the cloning of genomic DNA or chimeric cDNAs from different transcripts is extremely rare and allows for the recovery of complete singular genetic sequences.

    Library Design Preserves Full-Length cDNAs
    The goal was to develop a complete set of libraries representing the human transcriptome that aim to capture the rarest and least abundant species while enriching for the most complete transcripts, and maintaining the diversity of mRNA messages found in the original source material. For each group of cell lines or tissues, we synthesized first strand cDNA using an oligo dT priming approach followed by dG-tailing. After the dG-tailing procedure, we size fractionated the cDNA using electrophoresis in low melting point agarose gel to eliminate the possibility of cloning any residual molecules that may have been fragmented The cDNA produced was fractionated into two fractions: a low-sizing fraction, which contained cDNA molecules >800bp and high-sizing fraction containing cDNA >2000bp. Each fraction had different average insert lengths and was cloned separately into the pT3 vector to ensure representation of the longest transcripts without losing representation of smaller full-length molecules. Treating the class sizes separately eliminates competition at the cloning steps--ligation, transformation and growth naturally select for the smaller species--and therefore enhances for the representation of the longest transcripts. Thus, MegaMan™ Human Transcriptome Library due to its unique design and construction contains the longest and rarest human cDNAs.

    Primary Clones Minimizes Bias
    The MegaMan™ Human Transcriptome Library is a collection of primary clones that have never been amplified, thereby, preserving the representation of the rarest and most fragile mRNA sequences.
