Blunt End Cloning Kits

Blunt end cloning kits are available from a number of suppliers for the insertion of blunt-end PCR products into a plasmid vector. This cloning technique involves the insertion of DNA fragments with no overhanging bases at the 5’and 3’ ends into similarly blunt destination vectors. Blunt end cloning is often used when PCR products of interest contain common sticky end restriction sites, as this can cause undesired changes to the product if restriction endonucleases are used. Blunt end cloning kits generally combine the blunt end insert with the linear vector and DNA ligase, which then joins the genetic components into a single, completed plasmid. Selection markers, such as the ccdB gene, are often present to allow positive selection of recombinant vectors. Blunting kits, which remove incompatible 5' or 3' DNA overhangs, are also available.
