Description

Premixed, 2x concentrated High Fidelity PCR Master contains all necessary reagents (buffers, nucleotides and enzymes) needed for PCR. The mix contains a unique enzyme blend of Taq DNA Polymerase and Pwo DNA Polymerase[a thermostable enzyme with a proofreading (3'–5' exonuclease) activity]. Due to the inherent 3'–5' exonuclease proofreading activity of Tgo DNA Polymerase, the High Fidelity PCR Master increases the fidelity of DNA Synthesis approx. threefold compared to Taq DNA Polymerase. The High Fidelity PCR Master generates blunt-ended products and 3' single A-overhang products analogous to Taq DNA Polymerase. Note: Separate vials of PCR grade sterile water are included with the High Fidelity PCR Master

High Fidelity PCR Master is ideal for amplifying up to 5 kb targets from genomic DNA
High Fidelity PCR Master gives better results (higher yield and greater fidelity) than Taq DNA Polymerase alone in standard PCR reactions
High Fidelity PCR Master can be used instead of the Expand High Fidelity PCR System, or instead of single reagents

Operating parameters

Starting sample: up to 500 ng genomic or 10 ng plasmid DNA.
Temperature optimum (for elongation):
72°C (for <3 kb targets)
68°C (for >3 kb targets)
High Fidelity PCR Master accepts modified nucleotides like DIG-dUTP
