Optimizing Electroporation Parameters

Optimizing electroporation parameters

Other important parameters for optimizing electroporation.

Influence of the Cell Density

A cell density of 1 x 106 cells/ml is recommended for electroporation since the electrical field is still effective on the cells in this range. When high cell densities > 3 x 106 are used, homogeneous field conditions can no longer be guaranteed; i.e. the cells are no longer evenly exposed to the electrical field. This may lead to cell fusion and cause the transfection rate to decrease. However, using a cell density of < 1 x 106 cells/ml should have no negative effect on the transfection rate.
